摘要
目的探讨原发性肝细胞癌(HCC)中Gli2、Cyclin D1和p21的表达及其意义。方法采用组织芯片和免疫组织化学方法以及RT-PCR检测了44对HCC和相应癌旁肝组织中Gli2蛋白和mRNA的表达情况,应用Western blot检测了Cyclin D1、p21蛋白在两种组织中的表达差异。结果免疫组化染色与RT-PCR示Gli2在HCC组织中的阳性表达率为68.2%,而癌旁肝组织为38.6%,两者差异具有极显著性意义(P<0.01),且Gli2与HCC的组织分化和门静脉转移相关。Cyclin D1蛋白在HCC中的表达显著高于癌旁肝组织(P<0.05),且与Gli2的表达呈正相关;而p21蛋白在癌旁肝组织中的表达显著高于HCC,但与Gli2无明显相关性。结论Gli2过度激活可能通过上调Cyclin D1的表达而使肝癌细胞异常增殖,参与HCC的发生和发展。
Objective To study the expression of Gli2 in HCC and its relation with expression of Cyclin D1 and p21. Methods The protein and mRNA expression of Gli2 was detected by immunohistochemistry and RT-PCR and the protein expression of Cyclin D1 and p21 was detected by Western blot respectively in 44 cases of HCC tissues and corresponding adjacent-tumor liver tissues. Results The positive staining rate of Gli2 in HCC tissues was significantly higher than in the adjacent-tumor tissues (68.2% vs 38.6%, P〈0.01), and its expression in HCC was significantly correlated with the tumor differentiation and the tumor thrombus in portal vein. The expression of Cyclin D1 in HCC was higher than in adjacent-tumor tissues, but there was reverse to p21. There was a significant positive correlation between the expression of Gli2 with Cyclin D1, but not with p21. Conclusion The over-expression of Gli2 may play a crucial role in occurrence and progress of HCC by up-regulating Cyclin D1, which leads to the abnormal proliferation of HCC cells.
