摘要
目的观察白藜芦醇(resveratrol,Res)对纤维蛋白诱导的体外大鼠脑血管内皮细胞基质金属蛋白酶9(ma-trix metalloproteinases9,MMP9)高表达的影响。方法将大鼠脑血管内皮细胞分离后培养,培养液中加入1.0mg/ml纤维蛋白和不同浓度的白藜芦醇,通过RT-PCR检测脑血管内皮细胞中MMP9转录水平,应用明胶酶谱法(gelatin zy-mography)定量检测培养液中的MMP9水平。结果加入不同浓度(0、1、5、10、25和50μmol/L)的白藜芦醇24h后,25μmol/L和50μmol/L白藜芦醇组的培养液中MMP9水平显著降低(P<0.01);RT-PCR结果显示,25μmol/L和50μmol/L白藜芦醇组的大鼠脑血管内皮细胞中的MMP9显著下调(P<0.01)。结论白藜芦醇通过降低纤维蛋白导致的大鼠脑血管内皮细胞中MMP9的表达,在神经系统炎性病变中发挥抑制性保护作用。
Objective To investigate the effect of resveratrol on the matrix metalloproteinases9 (MMP9) protein and mRNA expression in the co-cultured rat brain vascular endothelial cells with fibrin. Methods The rat brain vascular endothelial cells were isolated, and co-cultured with 1.0 mg/ml fibrin and different concentrations of resveratrol (1-50 μmol/L). The expression of MMP9 mRNA was detected by using RT-PCR. The concentration of MMP9 in the culture was qunatitatively determined by gelatin zymography.Results After administration of different concentrations of resveratrol for 24 h, the MMP9 protein expression in the culture of 25 and 50 μmol/L vesveratrol-treated groups was significantly reduced (P〈0.01). RT-PCR revealed that in 25 μmol/L and 50 μmol/L resveratrol-treated groups the expression of MMP9 mRNA was significantly down-regulated in the rat brain vascular endothelial cells (P〈0.01). Conclusion Resveratrol plays the inhibitory protective role in neurologic inflammatory diseases by reducing the fibrin-induced MMP9 expression in rat brain vascular endothelial cells.
