摘要
目的研究成体骨骼肌肌源性干细胞(MDSCs)向神经细胞分化的潜能。方法从成年SD大鼠骨骼肌组织中培养出肌源性干细胞,采用bFGF、EGF诱导出神经球样细胞团,并检测nestin表达。再在贴壁培养条件下,用RA、BDGF诱导分化,并检测NSE、GFAP的表达。结果MDSCs能被诱导出神经球样细胞团,并表达nestin。这些细胞进一步诱导,部分细胞表现出神经元样或胶质细胞样形态,并表达NSE或GFAP。结论MDSCs能向神经细胞样细胞分化,有望用于神经系统疾病的治疗。
Objective To study the differentiation of skeleton muscle-derived stem cells (MDSCs) into neural cells. Methods MDSCs were cultured from adult SD rat skeleton muscle. Nestin molecule of these cells was inspected after they were cultured in non-adherent condition with bFGF and EGF. The expression of NSE and GFAP was detected in these cells after they were induced in adherent cultural condition with RA and BDNF. Results Neurosphere-like cell agglomerates were gained after MDSCs were induced by bFGF and EGF. In the adherent cultural condition, some cells of these agglomerates turned to neuron-like cells or glial-like cells after RA and BDGF were added. Conclusion MDSCs show their potential of transdifferentiation into neural cells.
