具有控释性能的可注射牙槽骨修复材料中β-磷酸三钙的体外细胞毒性研究 被引量:9

摘要 目的利用体外细胞培养技术,对具有控释性能的可注射牙槽骨修复材料中β-磷酸三钙的细胞毒性进行研究。方法采用体外培养的小鼠成纤维细胞(L-929),在培养液中分别添加不同浓度的β-磷酸三钙浸提液(原液及2、4、8、16倍稀释液),于接种后72h通过MTT法显色,在酶标仪490nm波长处测定吸光度值,计算相对增殖率(RGR),对β-磷酸三钙的细胞毒性进行评价。结果各组的RGR值均≥80%,各浓度组之间差异无显著性意义(P〉0.05),提示L-929细胞增殖与β-磷酸三钙浸提液的浓度无明显相关,不同浓度浸提液的细胞毒性为0-1级。结论具有控释性能的可注射牙槽骨修复材料中β-磷酸三钙对细胞的生长和增殖无明显抑制作用,无明显的细胞毒性。 Objective To investigate cytotoxicity of β-tricalcium phosphate in a novel injectable alveolar replacement material as a drug delivery by MTT assay. Methods L-929 mouse fibroblasts were cultured with different concentrations of the extracted liquid of β-tricalcium phosphate (by the 2, 4, 8, 16 double dilution) in vitro, and MTT assay was performed to evaluate the cell relative proliferation rate at 72 h. Results MTT assay revealed that there was no significant difference in cytotoxicity among the different concentrations groups of extracted liquid of β-tricalcium phosphate (P〉0.05). The cytotoxicity of β-tricalcium phosphate was between 0 and 1 grade according to the test standard of USP, suggesting that the material had no cytotoxicity.Conclusion The β-tricalcium phosphate possesses ideal cellular compatibility.
