摘要
目的:测定陕西产茜草中1,3,6-trihydroxy-2-methylanthraquinone-3-O-3-[3-O-acetyl-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside]的含量。方法:选用Purospher star RP-C18色谱柱(4.6 mm×250 mm,5μm,Merck公司);流动相为甲醇-水-四氢呋喃(65∶34.7∶0.3),流速为1.0 ml/min,检测波长为276 nm,柱温为25℃,外标法定量分析。结果:该方法在0.020μg^0.160μg范围内线性关系良好,r=0.9999,平均回收率101.4,RSD=2.0%(n=6),测定结果为0.053%。结论:该法测定茜草中1,3,6-trihydroxy-2-methylanthraquinone-3-O-3-[3-O-acetyl-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside]含量灵敏、准确、重现性好。
Objective: To determine 1,3, 6-trihydroxy-2-methylanthraquinone-3-O-3-[ 3-O-acetyl-α-L-rhamnopyranosyl-( 1 →2)-[3-D-glucopyranoside] in Rubia. cordifolia from Shanxi Province by RP-HPLC. Methods: The HPLC system consisted of Purospher star RP-C18 column (4.6 mm × 250 mm, 5 μm, Merck) at 25℃, MeOH-H2O-tetrabydropyrane (65: 34.7: 0.3) mixture as mobile phase, detection wavelength at 276 nm, flow rate of 1.0 ml/min. Results: The linearity was found in the range of 0.020-0. 160 ug, r =0.9999, the average recovery'was 101.4%, RSD was 2.0%(n = 6). The contents of 1, 3, 6-trihydroxy-2-methylanthraquinone-3-O-3-[ 3-O-acetyl-c^-L-rhamnopyranosyl-( 1 →2)-[3-D- glucopyranoside] in Rubia. cordifolia from Shanxi province was 0. 053%. Conclusion: The method is reliable and suitable for determination of contents of 1,3, 6-trihydroxy-2-methylanthraquinone73-O-3-[ 3-O-acetyl-α-L-rhamnopyranosyl- (1→2)-[3-D-glucopyranoside] in Rubia. cordifolia from Shanxi Province.
