摘要
目的探讨水通道蛋白-5(aquaporin5,AQP5)在变应性鼻炎(allergic rhinitis,AR)患者鼻黏膜中的表达及意义。方法应用免疫组织化学技术检测32例变应性鼻炎患者(变应性鼻炎组)下鼻甲黏膜和25例行鼻中隔偏曲矫正术的无变应性鼻炎患者(对照组)下鼻甲黏膜组织中AQP5表达情况,分析AQP5表达与变应性鼻炎严重程度间的关系。结果①对照组鼻黏膜HE染色呈轻度炎症反应,AQP5表达较弱;AQP5染色阳性细胞很少且散在分布于鼻黏膜上皮下细胞。②变应性鼻炎组患者鼻黏膜HE染色表现为明显的腺体增生、炎症反应和嗜酸粒细胞浸润;免疫组织化学染色显示鼻黏膜组织中大量AQP5阳性细胞,主要分布在腺上皮细胞、导管上皮细胞、纤毛上皮细胞以及鼻黏膜血管的内皮细胞的胞膜和胞质中,与对照组比较AQP5表达阳性细胞数差异具有统计学意义(P<0.01)。③变应性鼻炎组AQP5的表达与变应性鼻炎严重程度密切相关(r=0.819,P<0.001)。结论AQP5在变应性鼻炎鼻黏膜中高表达,能客观反映免疫或炎症反应的程度,AQP5的高表达与腺体过度分泌密切相关。对AQP5的研究开发可能成为为变应性鼻炎诊断评分系统的补充及新的基因治疗效靶。
[Objective] To evaluate the expression and its significance of aquaporin 5 (AQPS) in nasal mucous of allergic rhinitis (AR). [Methods] The expression of AQP5 by immunchistochemical staining was examined in the inferior turbinate tissues from 32 allergic rhinitis patients and from 25 non-allergic patients underwent nasal septum operation, the relationship between AQP5 and severity of AR was analysed. [Results] ① HE staining showed that there was mild inflammation reaction, exporession of AQP5 was lowe and merely located within epithelium and subepithelium sparsely in the contral group. ② In the AR group, HE staining showed inflammation reaction was obvious,a large quantity of glands hyperplasia and acidophils soaked in nasal mucosa. AQP5 was strongly expressed and its expression mainly in the membrane and cytoplasm of the epithelium in the glands, ducts, cilia, and the blood vessel endothelial cell, the numbers of positive AQP5 cell had significant deference between the AR group and the control group (P 〈0.01). ③The numbers of positive AQP5 cell were correlated with the severity of AR (r = 0.819, P 〈0.001). [Conclusions] AQP5 is expressed highly in nasal mucous of AR, it reflects the degree of immunology and inflammation, the high expression of AQP5 has a close relationship with the hypersecretion of glands in allergic rhinitis. The exploration of its potential role can provide a diagnostic tool and a target for gene therapy for AR.
