摘要
通过筛选,得到7%的硫酸锌溶液与无水乙醇以10:1(体积比)的混合溶液能使灵芝孢子更好的悬浮。利用该悬浮体系,辅以超声60 min和涡旋处理30 s,通过血球计数板对不同质量的孢子粉进行计数,建立未破壁灵芝孢子粉质量和孢子数之间的标准曲线,对同批未知破壁率的破壁孢子粉的破壁率进行检测,并人为混合7个标准破壁率的破壁孢子粉对该方法的准确度和精密度进行试验。结果表明:在该悬浮体系下,灵芝孢子粉在0.01~0.05 g/25 mL与孢子数线性关系良好。
The selection experiment showed that the mixed solution of 7% zinc sulfate solution and anhydrous ethanol at volumetric ratio = 10 : 1 could make the spores suspend well. With this solution the spores of different masses were counted by a hemacytometer after 60-minute ultrasonic wave and 30-second eddy treatment,and so a standard curve was set up between the mass and the number of intact spores. A broken sporoderm rate of the spores' product was determined according to the standard curve. Test for 7 standard broken sporoderm rates of the artificially mixed spores showed that there was a good linear relationshio between the spores number and the spores of 0. 01-0.05 g/25 mL.
