摘要
目的评价反义LeETR1转基因番茄(ETR1)向动物肠道菌群和子代动物转移外源基因的能力及对小鼠生殖发育的影响。方法SD大鼠和ICR小鼠连续饲喂ETR1、非转基因番茄(B1)和蒸馏水30 d后,分别进行大鼠盲肠菌群培养物的抗生素耐药性试验及小鼠生殖毒性试验,提取耐药阳性菌、亲代和子代小鼠的基因组进行外源基因特异性聚合酶链式反应法(PCR)。结果肠道菌的耐药性和小鼠生殖发育情况无明显改变,在耐药菌和两代小鼠体内均未发现由受试物引入的外源基因。结论ETR1对小鼠无生殖毒性,其携带的外源基因不能向动物体细胞、肠道菌及子代动物转移。
Objective To assessment the effect of anti-LeETR1 genetically modified tomato (ETR1) on reproductive- development toxicity and transferability of transgenes. Methods SD rats were sacrificed after being administered with ETR1, common tomato (B1) and water for 30 days and the bacteria in cecum were picked out for anti-antibiotics analysis. ICR mice were used for reproductive toxicity after 30 days administration. Genomic DNA of the tolerant bacteria and two generations of the mice was extracted and amplified by transgene-specific PCR. Results There is no obvious difference among the groups on the ability of tolerance and reproductive profile. In the tolerant bacteria and two generations of the mice, there is no tested transgene introduced by ETR1. Conclusion ETRI had no reproductive toxicity effect on mice and couldn't deliver the transgene to animal's somatic cell, cecum bacteria and their offspring.
