摘要
目的:检测一氧化氮合酶(NOS)抑制剂非对称性二甲基精氨酸(ADMA)对原发性高血压(EH)患者外周血单核细胞趋化活性的影响。方法:收集42例EH患者(EH组)和40例门诊健康体检者(对照组)。检测血浆中ADMA和NO水平、血管假性血友病因子(vWF)、白细胞介素8(IL-8)和肿瘤坏死因子(TNF)-α浓度,并观察ADMA对外周血单核细胞IL-8受体CXCR2 mRNA表达的影响。结果:与对照组比较,EH患者血浆中AD-MA和vWF水平升高、NO生成减少(P<0.05),同时血中IL-8和TNF-α水平显著升高(P<0.05)。EH患者分离的外周血单核细胞CXCR2 mRNA表达明显高于对照组,外源性的ADMA 30μmol/L孵育4 h后可进一步上调CXCR2 mRNA的表达(P<0.05)。结论:EH患者血中升高的ADMA可能增加外周血单核细胞的趋化活性,促进动脉粥样硬化的发生。
Objective:To determine the role of endogenous nitric oxide synthase (NOS) inhibitor such as asymmetric dimethylarginine (ADMA) in ehemokine activity of peripheral blood mononuelear cells (PBMCs) in individuals with primary hypertension. Method: Fourty-two patients with essential hypertension and 40 healthy controls in the out-patient clinic and admission department of the hospital were enrolled in the present study from Aug 2005 to May 2006. Plasma levels of ADMA, NO, vonwillebrand factor (vWF), interleukin (IL-8) and tumor necrosis factor α (TNF-α) were measured and the effect of ADMA on chemokine receptors CXCR2 mRNA expression in PBMCs was also determined. Result.. Compared with controls, the levels of ADMA and vWF were markedly elevated and the levels of NO were decreased (P〈0.05), companied with elevated levels of IL-8 and TNF-α in hypertensive patients (P〈0.05). The expression of CXCR2 mRNA in separated PBMC was significantly higher in individuals with hypertension than that in controls, which was further upregulated by incubation with ADMA (30 μmol/L, 4 h). Conclusion: Elevated ADMA level in hypertensive patients may facilitate monocytic chemokine activity, then to promote the development of atherosclerosis.
