摘要
目的探讨线粒体氯通道蛋白/CLIC4在局灶性脑缺血损伤中的作用。方法应用线栓法建立大鼠大脑中动脉阻塞(MCAO)模型。RT-PCR检测脑皮质CLIC4和诱导型一氧化氮合酶(iNOS)的mRNA表达;Western印迹方法检测脑皮质CLIC4的蛋白水平。结果与假手术组相比,缺血模型大脑皮质缺血侧CLIC4及iNOS的mRNA明显增高,CLIC4蛋白水平亦增加,而缺血模型正常侧大脑皮质CLIC4 mRNA及蛋白表达均降低。结论CLIC4可能参与脑缺血引起的神经细胞损伤。
Objective To explore the effect of mitochondria chloride channel (CLICA)on rat cortex with middle cerebral artery occlusion (MCAO). Methods The focal cerebral ischemia model was established by MCAO. CLIC4 mRNA and iNOS mRNA expression were assayed by RT-PCR and CLIC4 protein expression was measured by Western blotting. Results CLIC4 and iNOS mRNA expressions and CLIC4 protein expressions in ischemia side of MCAO group were obviously higher than those of sham group, but CLIC4 mRNA and protein expression in normal side in MCAO group were obviously lower than those of sham group. Conclusious CLIC4 might be involved in brain injury induced by focal cerebral ischemia.
