摘要
目的观察骨质疏松小鼠SAMP6主动脉病变的特点。方法以SAMP6小鼠及其同源对照鼠SAMR1为模型,采用放免法测定血清骨钙素(BGP)水平;原子吸收法测定主动脉等组织的钙含量;HE和von Kossa染色法观察主动脉病变情况。结果与SAMR1组比较,SAMP6组血清BGP升高;血钙、骨钙浓度较低;而尿钙、主动脉钙含量较高;主动脉中膜平滑肌细胞数量少,排列紊乱;中弹力板多处疏松、断裂;中膜弹性纤维间有钙沉积颗粒。结论SAMP6小鼠伴衰老有主动脉形态结构异常及中膜轻度钙化特征,可作为钙化矛盾模型加以利用。
Objective To observe the pathological changes of the aorta in SAMP6 mice. Methods The serum BGP level of SAMP6 and SAMR1 mice were determined using radio immunoassay, and the calcium contents in several tissues were assayed with atomic absorption spectrometer. The sections were stained by HE and von Kossa methods to explore the morphological features of aortic lesions. Results Compared to SAMR1 group,the calcium content in serum and femur of SAMP6 decreased, whereas the serum BGP level,the calcium concentration in urine and aorta increased obviously. Under light microscopy ,the smooth muscle ceils in tunica media of SAMP6 decreased in amount and arranged disorderly, and the elastic lamella were severely disorganized with areas of complete disappearance of the elastic fiber. Calcium deposits were detected within the elastin fibers of tunica media. Conclusions Slight calcification is detected in the tunica media of SAMP6, which suggests that the mouse model would be useful for further investigation in calcification paradox.
