摘要
以中国鹅掌楸、北美鹅掌楸、杂种鹅掌楸的幼芽为材料,利用Trizol、CTAB、SDS和异硫氰酸胍法提取鹅掌楸植物总RNA,结果表明,(1)用Trizol法提取的中国鹅掌楸、北美鹅掌楸、杂种鹅掌楸的RNA具有28S、18S、5S rRNA 3条较清晰的条带,很少有降解,其完整性很好;(2)用Trizol法获得的3种鹅掌楸RNAD260/280值为1.724-1.801,D260/230值为2.154-2.341,说明其纯度也很好;(3)比较4种方法对提取鹅掌楸植物总RNA的得率发现,用Trizol法提取的RNA得率为219.2-256.7μg.g-1,是其它2种方法的1-9倍。因此,Trizol法可获得较高质量与产量的鹅掌楸植物总RNA,可满足下一步的研究。
Total RNA was isolated from the plumules of Liriodendron chinense, L. tulipifera and their hybrids (L. chinense × L tulipifera) with the methods of Trizol, CTAB, SDS and guanidinium isothiocyanate. The results were as follows : ( 1 ) The total RNA isolated from all experimental materials by the method of Trizol had three clear bands-28S rRNA, 18S rRNA, 5S rRNA. In addition, the total RNA was undegraded and highly-complete. (2) The D260/280 absorbance ratio of all RNA from experimental materials by the method of Trizol ranged from 1.724 to 1.801. And their value of D260/230 ranged from 2,159 to 2.341. The results demonstrated the RNA was highly pure. (3) Comparing the yields of RNA isolated by the four methods, RNA obtained by the method of Trizol had highest productivity 219.2 -256.2 μg · g^-1. This was between one and nine times as much as the other two methods. So, the RNA obtained by the method of Trizol was of high quality, high yields, and suitable for all downstream applications.
