摘要
腺病毒载体具有高效转染和低细胞毒性的特点,已成为基因治疗研究中应用最广泛的载体之一。作为最新一代腺病毒载体,第三代腺病毒载体由于其生产效率低下,难以生产临床使用的治疗剂量,所以一直没有临床实验的结果报道。为了提高腺病毒的生产效率,文章研究E1区对腺病毒在CA293细胞中生产能力的影响,结果发现E1区基因拷贝数和蛋白表达与腺病毒的产量呈正相关,而不影响病毒DNA在CA293中的复制能力,推测E1区影响病毒DNA复制后的装配和释放过程,为改进腺病毒的生产方法提供理论基础。
Adenovirus vectors have the characteristics of high transfection efficiency and low cytotoxicity, and they have become most widely used vectors for gene therapy. As the last generation adenovirus vector, the third generation can hardly increase to therapeutic dose for its low productivity, and thus no clinical report of it has been published by far. In order to improve adenovirus productivity, the authors study the effect of E1 region on adenoviral production capacity in CA293 cell line. It is observed that E1 region gene copies and protein expression are positively related to adenoviral production while they have no influence on viral DNA replication in CA293, indicating that E1 region affects the process of virus assembly and release. The research provides the theoretical basis for advances in procedures of adenoviral production.
