摘要
目的:建立冰蟾乳膏中马钱子碱和士的宁的含量测定方法。方法:采用DiamonsilTMC18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.015 mol/L庚烷磺酸钠与0.02 mol/L磷酸二氢钾等量混合溶液(含0.1%三乙胺,用10%磷酸调节pH值至3.0)(20∶80),流速:1.0 mL/min,检测波长:256 nm。结果:马钱子碱在2.4-24 mg/L范围内(r=0.999 7)、士的宁在2.95-29.5 mg/L范围内(r=0.999 9)线性关系良好,其平均加样回收率分别为98.3%和97.9%,RSD分别为1.54%和2.20%(n=5)。结论:本法快速、准确、重复性好,可作为冰蟾乳膏的质量控制方法。
Objective: To establish a method for the determination of brucine and strychnine in BingChan cream by RP-HPLC. Methods: A DiamonsilTM C18 column(250 mm ×4.6 mm, 5 um) was used, the mobile phase was acetonitrile-equivalent mixture consisted of 0.02 mol/L KH2PO4 and 0. 015 mol/L sodium heptanesulfonate with 0.1% triethylamine (pH 3.0) (20: 80), the flow rate was 1.0 mL/ min, and the detection wavelength was 256 nm. Results: The linear range of brucine and strychnine was 2.4-24 mg/L (r =0.9997) and 2.95-29.5 mg/L (r = 0. 999 9), respectively, and the mean recovery was 98.3% ( RSD = 1.54%) and 97.9% ( RSD = 2.20%), respectively. Conclusion: The method is simple, accurate and reliable for the quality control of BingChan cream.
