摘要 目的:制备苦参碱微乳透皮给药系统,对其理化性质及透皮行为进行考察。方法:以苦参碱、磷脂、聚氧乙烯辛基苯基醚、肉豆蔻酸异丙酯和去离子水为主要成分,水相溶液逐滴加入油相的方法,制备苦参碱微乳,考察其外观形态、粒径大小及分布,采用Valia-Chien扩散池考察苦参碱微乳的体外透皮过程。结果:制得的苦参碱微乳为淡黄色透明液体,平均粒径120 nm,外观圆整均匀,24 h的累积释放量为(1 056.1±74.9)μg/cm2。结论:苦参碱微乳具有良好的透皮效果,为苦参碱的临床应用提供了一种新型制剂。 Objective: To prepare matrine loaded microemulsion as transdermal delivery system and study its physicochemical properties and skin permeability. Methods: Traditional water dropwise titration method was adopted to prepare microemulsion using matrine, phospholipid, Triton X-100 (OP) ,isopropyl myristate and deionized water as main ingredients. The morphology and grain size distribution of microemulsion were studied. Skin permeability test was performed using Valia-Chien cell. Results:The microemulsion was yellow in color and its grains were round,with mean grain size at 120 nm in diameter. Cumulative permeation quantity in 24 h was (1 056.1 ± 74.9) μg/cm^2. Conclusion: Matrine loaded microemulsion has good permeability. It is a practical preparation of matrine for clinical use.
