异基因外周造血干细胞移植术后CD4^+CD25^+调节性T细胞相关性研究

摘要 为研究异基因外周血干细胞移植(Allogeneic peripheral blood stem cell transplantation,Allo-PBSCT)患者外周血CD4+CD25+调节性T细胞(CD4+CD25+regulatory Tcells,CD4+CD25+Tregs)及其表达的CD28,CD152的变化,选取行ALLO-PBSCT的51例血液病恶性肿瘤患者在移植前,植入,选取+30d,+60d,+120d,+360d 6个时间点,采用流式细胞术动态监测外周血CD4+CD25+Tregs数量及其表达CD28,CD152的百分含量。Allo-PBSCT术后,CD4+CD25+Tregs数量及其表达CD28均先降后升又再次回落到接近植入前水平,二者呈正相关,CD152的表达与移植前比较,在各时间点的表达水平很低,且均无显著性差异。Allo-PBSCT过程中,CD28的表达决定着CD4+CD25+Tregs的稳态,Allo-PBSCT患者外周血CD4+CD25+Tregs的数量增加,但未被激活。 To investigate the variation of CD4^+CD25^+ regulatory T cells and CD28 ,CD152 expressed by them in allogeneie peripheral blood stem cell transplantation( Allogeneie peripheral blood stem cell transplantation ,Allo - PBSCT) patients'peripheral blood, the amount of CD4^+CD25^+ Tregs and the percentages of CD28,CD152 expressed by them in the peripheral blood of 51 patients with hematological maglignancies were detected by flow eytometry in the course of Allo - PBSCT on pretransplantation, during transplantation, + day 30, + day 60, + day 120 and + day 360 respectively. In the course of Allo - PBSCT the amount of CD4^+CD25^+ Tregs and CD28 expressed by them declined followed by increase and then fell again to approach to the pretransplantation level. They were in positive correlation with each other. CD152 was expressed at very low level which had no significant difference among the values on different time in the course of Allo - PBSCT. In the course of Allo - PBSCT the stable anaotmt of CD4^+CD25^+ Tregs was determined by CD28 expressed by them. The CD4 + CD25 +Tregs in Allo- PBSCT patients'peripheral blood increased but werent activated.
