摘要
目的观察脂联素对糖尿病动物模型的糖脂代谢影响。方法高糖高脂饲养小香猪6个月,构建糖尿病动物模型。将糖尿病小香猪随机分为两组:注射生理盐水组(正常组)、注射脂联素组(脂联素组),每组5只。体外表达重组脂联素,一次性腹腔注射脂联素,检测给药后血糖、胰岛素的变化。结果一次性注射脂联素后,体内血糖浓度显著降低。口服糖耐量(OGTT)实验表明脂联素改善了胰岛素敏感性。结论重组脂联素具有生物学活性,且脂联素能降低糖尿病动物的高血糖状况,改善胰岛素敏感性,为人类糖尿病的治疗提供一定的理论基础。
Objective To investigate the effects of recombinant human adiponectin on metabolization of diabetic swines. Methods Ten chinese Guizhou swines were fed by high fat/high-sucrose fodder, and observed for 6 months. Then the ten diabetic swines were divided in to two groups: nolmal group and adiponectin group. We assessed the effects of recombinant adiponectin by detecting the change of plasma glucose levels and evaluating the insulin sensitivity. Plasma glucose was determined by commercially available enzymatic method test kits, and insulin sensitivity was evaluated by oral glucose tolerance test (OGTr). Results A single injection in diabetic swines of purified recombinant human adiponectin triggered a transient decrease in basal glucose levels (P〈0.05). Moreover, after adiponectin injection, swines also showed to improve insulin sensitivity compared with the control (P〈0.05). Conclusion Adiponectin might have potential as a glucose-lowering agent and as a therapeutic for metabolic disease.
