摘要
采用农杆菌介导法将高赖氨酸基因导入脆茎粳稻诱导的愈伤组织,经潮霉素抗性筛选,得到可育的再生植株,经PCR检测,表明该基因已整合到脆茎粳稻的基因组中。对部分T2代和花药培养TH1代纯合株系进行研究,结果显示,与非转基因对照相比,在生育期相仿条件下,可以选到生物量和穗重无显著差异,收获指数有提高趋势的转基因水稻;转基因水稻秸秆赖氨酸和粗蛋白含量都有不同程度的提高,最高含量分别为0.350%和7.27%,但转基因水稻秸秆的其他氨基酸含量的变化趋势并不一致。选出了2个株系可能适合作秸秆饲用化的育种材料。
The lysine-rich protein gene from winged bean (WBLRP) and hygromycin phosphotransferase gene through a binary expression vector were introduced into brittle culm rice by Agrobacterium -mediated transformation. PCR analysis indicated that the lysine-rich protein gene had been integrated into the nuclear genome of a fertile transgenic rice. The harvest index, lysine content and crude protein content of culms for the T2 and TH1 transgenic pure lines were studied. The results showed that there are no significant differences in biomass, panicle weight per plant and there is a slight increse in harvest index. The crude protein and lysine contents in culms of the transgenic lines were higher than the control, the highest reached 7.27 % and 0.35 % respectively. However, the changes of the contents of methionine and other amino acids in culms were not consistent comparing with that of lysine. Two lines were selected as potential breeding materials for straw feeding purpose. The application of the transgenic brittle culm rice in rice production was also discussed in the paper.
