摘要
运用Plackett-Burman试验设计筛选对黑曲霉XY-7生产木聚糖酶有显著影响的因素。在研究的20个因素中,氯化钙、木糖、蔗糖、棉子糖和硫酸亚铁5种因素明显抑制黑曲霉XY-7的产酶水平,为产酶负影响因子;硝酸钠、酵母粉、尿素、碳酸钠、硫酸镁、蛋白胨和硫酸铵7种因素强烈诱导A.nigerXY-7的产酶水平,为产酶正影响因子。氮源中尤以硝酸钠诱导效果最显著,其次是酵母粉和尿素。
Nutritional requirements was different for xylanase production of various strains. Plackett-Bunnan experimental design was applied to evaluate the nutritional requirements of Aspergillus niger XY-7 xylanase production. Among studied 20 factors, CaCl2, xylose, sucrose, raffinose and FeSO4 repressed the xylanase production significantly, and NaNO3, yeast extract, urea, Na2CO3, MgSO4, peptone and (NH4)2SO4 induced the xylanase production strongly. NaNO3 was identified as the best nitrogen source for Aspergillus niger XY-7 xylanase production, while yeast extract and urea served as secondary best nitrogen source.
