摘要
目的:制备豚鼠变应性鼻炎(AR)动物模型,通过干预性处理研究内源性硫化氢(H2S)及其限速酶胱硫醚-γ-裂解酶(CSE)在AR中的变化及意义。方法:将24只豚鼠随机分为4组,分别为卵清蛋白致敏豚鼠变应性动物模型组(致敏组)、NaHS处理组及CSE抑制剂炔丙基甘氨(PPG)干预组(PPG组),同时以生理盐水处理豚鼠作为对照组,分别测定各组动物鼻腔灌洗液eotaxin、血浆H2S浓度,采用实时荧光定量RT-PCR法测定鼻黏膜中CSE的表达。结果:豚鼠致敏后eotaxin含量明显增加,而血浆H2S浓度及鼻黏膜CSE的表达明显低于对照组(P<0.05),而干预处理的NaHS组与致敏组相比,eotaxin相应减少(P<0.05),而H2S浓度及CSE的表达量增加(P<0.05),2组间差异有统计学意义,而经PGG干预处理后,eotaxin含量则增加,而H2S浓度及CSE的表达量降低,2组间差异有统计学意义(P<0.05),其中H2S含量与CSE的表达呈线相关。结论:内源性H2S参与AR的炎症过程,其含量主要受限速酶CSE的调控。
Objective:To study the change of endogenous hydrogen sulfide (hydrogen sulfide, H2S) and its rate-limiting enzyme Cystathionine-γ-lyase (CSE) in allergic rhinitis through guinea pigs with intervention treatment. Method:Twenty-four guinea pigs were divide into 4 groups at random, one group were models of allergic rhinitis (AR) which were established by using ovalbumin, the second group were treated with Naris after sensitized, the third group were treated with Propargylglycine (PPG) which was suppression of CSE after sensitized, and the last group were treated with saline for control. The concentration of eotaxin of nasal lavage and H2S in plasma were recorded, and then the expression of CSE in nasal mucosa was determined by real-time fluorescence RT-PCR. Resalt:The concentration of eotaxin in nasal lavage of sensitized group were higher than those of control (P〈0.01), and concentration of H2 S in plasma and expression of CSE in nasal mucosa were lower than control (P〈0.05). The concentration of eotaxin decreased when treated with Naris and increased when treated with PGG (P〈0.05). Level of H2S in plasma and expression of CSE increased when treated with Naris and decreased when treated with PGG (P〈0.05), and the level of H2 S was positive linear correlate with the expression of CSE. Conclusion:Endogenous H2S perhalps plays a significant role in the pathogenesis of allergic rhinitis, and it was mainly regulated by CSE.
