摘要
目的探讨线粒体DNA(mtDNA)Cyt-b和血管内皮生长因子(VEGF)mRNA的表达在乳腺癌发生、发展过程中的意义。方法采用逆转录-聚合酶链反应(RT-PCR),分别对19位乳腺癌患者的19例乳腺癌组织标本和19例癌旁组织标本的mtDNA Cyt-b、VEGF mRNA的表达进行检测(β-actin作为定量标准物),观察它们在乳腺癌组织和癌旁正常组织中的表达差异以及与临床病理参数的关系。结果19位乳腺癌患者中,乳腺癌组织mtDNACyt-b和VEGF mRNA的表达均高于癌旁组织,两组比较均有明显差异(P<0.01),而乳腺癌组织中VEGF的两个异构体VEGF145和VEGF189的mRNA表达高于癌旁组织,两组比较均有差异(P<0.05),并且mtDNA Cyt-b与VEGFmRNA的表达呈正相关性(P<0.01),与VEGF145、VEGF189的mRNA表达均呈正相关性(P<0.05);淋巴结转移组mtDNA Cyt-b和VEGF mRNA的表达较无淋巴结转移组高,两组比较均有差异(P<0.05);TNM分期Ⅰ期mtDNA Cyt-b和VEGF mRNA的表达较Ⅱ期低,两组比较均有差异(P<0.05);Ⅰ期与Ⅲ期比较两组mtDNA Cyt-b mRNA的表达有明显差异(P<0.01),而VEGF mRNA的表达有差异(P<0.05)。结论乳腺癌中mtDNA Cyt-b和VEGF mRNA的表达上调,并且在肿瘤的能量传递过程起着一定作用。
Objective To study the significance of the expression of mitochondrial DNA Cyt-b and vascular endothelial growth factor mRNA in human breast cancer. Methods Reversed-transcription polymerase chain reaction(RT-PCR)was applied to detect the expressions of mtDNA Cyt-b and VEGF in 19 samples of breast cancer tissue and 19 samples of peritumoral tissues. β-actin served as a quantitative standard marker. The different expressions in breast cancer tissues and paracancerous tissues were compared and their relations with clinical pathology parameters were analyzed. Results The expressions of mtDNA Cyt-b and VEGF mRNA in 19 samples of breast cancer tissues were higher than those in paracancerous tissues, with a significant difference between the two groups ( P 〈 0.01 ). The expression of mtDNA Cyt-b was positively correlated with the expression of VEGF mRNA ( P 〈 0.01) and the expression of VEGF145 and VEGF189 mRNA (all P 〈 0.05). The expression of VEGF145 and VEGF189 mRNA, which were the isomerides of VEGF, was higher in breast cancer tissues than in paracancerous tissues, with a difference between the two groups ( P 〈 0.05). The expression of mtDNA Cyt-b and VEGF mRNA in lymph node metastasis group was higher than that of non-lymph node metastasis group, with a difference between the two groups ( P 〈 0.05). The expressions of mtDNA Cyt-b and VEGF mRNA in phase Ⅰ of TNM stage were lower than those in phase Ⅱ , with a difference between the two groups ( P 〈 0.05). There was a significant difference in the expressions of mtDNA Cyt-b mRNA between the two groups in phase Ⅰ and phase Ⅲ (P 〈 0.01 ), and there was difference in the expression of VEGF mRNA ( P 〈 0.05) . Conclusion The expressions of mtDNA Cyt-b and VEGF mRNA in breast cancer are upregulated. They play an important role in the process of transmitting energy.
