RNA干扰胰岛素样生长因子结合蛋白2基因治疗胶质瘤的体外实验研究

摘要 目的研究与胰岛素样生长因子结合蛋白2(IGF-BP2)对胶质瘤的抑制作用。方法化学合成3条针对IGF-BP2的siRNA和1条阴性对照siRNA,采用小分子干扰RNA技术使胶质瘤U251细胞的IGF-BP2基因沉默。通过逆转录PCR和免疫组化的方法检测IGF-BP2的表达,用MTT法检测干扰后U251细胞的增殖活性变化。结果逆转录PCR检测表明转染后24hIGF-BP2的mRNA表达明显降低,空白对照、阴性对照、脂质体、siRNA1、siRNA2、siRNA3组中IGF-BP2mRNA水平分别为:0.88±0.02,0.87±0.03,0.84±0.02,0.65±0.03,0.55±0.04和0.64±0.02。48h分别为0.93±0.04,0.90±0.04,0.93±0.03,0.76±0.04,0.58±0.03和0.75±0.02,72h为0.95±0.04,0.95±0.03,0.94±0.02,0.78±0.05,0.60±0.02和0.76±0.03。3条siRNA中siRNA2作用最显著。免疫组织化学方法检测表明IGF-BP2的蛋白表达阳性率也由92.3%明显降低至24h的15%,至48h为52.5%,仍低于正常胶质瘤细胞。RNA干扰后胶质瘤细胞的MTT值虽在缓慢升高,于24h相对于对照组差异无统计学意义,但至48h和72h均低于对照组(P<0.05)。结论理想的IGF-BP2小分子干扰RNA序列能够抑制该基因的mRNA和蛋白的表达并能抑制胶质瘤细胞的增殖活性。
