摘要
目的探讨不同亚群树突状细胞(DC)对初始型T辅助细胞(Th0)分化的影响。方法分离实验性自身免疫性脑脊髓炎(EAE)易感性BALB/c小鼠脾脏的DC前体细胞,与髓鞘碱性蛋白(MBP)及重组小鼠白细胞介素-4(IL-4)I、L-3共培养,然后取上述培养的DC与单个核细胞(mononuclear cell,MC)培养,测定培养液上清液中干扰素-γ(IFN-γ)和IL-4水平。结果DC与MC共培养经IL-4刺激后其上清液中IFN-γ水平[(23.8561±3.2675)pg/mL]较IL-3刺激组[(20.8422±3.5225)pg/mL]及对照组[(20.2845±3.1043)pg/mL]明显增高(P<0.05);经IL-3刺激后其上清液中IL-4水平[(10.9457±1.9180)pg/mL]较IL-4刺激组[(9.1826±1.5361)pg/mL]及对照组[(9.2046±1.3261)pg/mL]明显增高(P<0.05)。结论不同亚群DC对Th0细胞分化的影响不同,提示可通过调控DC的分化方向调控Th0细胞的分化。
Objective To investigate the effect of different DC subpopulations on the differentiation of Th0 cells. Methods The precursor DCs, separated from the spleen of BALB/c mice, were co-cultured with MBP and recombinant mice IL-4 and IL-3 respectively. And the precursors were susceptible to EAE. The cultivated DCs were then co-cultured with mononuclear cells (MC), and the levels of IFN-γ and IL-4 in supernants were detected. Results The level of IFN-γ in the supernants of the DCs co-cultured with MC stimulated by IL-4 [(23. 8561±3. 2675)pg/mL] was significantly higher than that of the group stimulated by IL-3[(20. 8422 ± 3. 5225) pg/mL] and the normal control [(20. 2845 ± 3. 1043)pg/mL], while the level of IL-4 [(10. 9457 ±1. 9180)pg/mL] induced by IL-3 was significantly higher than that of the group stimulated by IL-4[(9. 1826± 1. 5361)pg/mL] and the normal control[(9. 2046± 1. 3261)pg/mL]. Conelusions The different subpopulations of DCs had different effects on the differentiation of Th0, indicating that the diffenentiation of the Th0 subsets could be regulated by modulating the DC.
