摘要
以黑芝麻无菌苗的下胚轴、幼根、子叶为外植体,以MS+2,4-D1.5mg/L+ZT0.5mg/L为愈伤组织诱导培养基;MS+6-BA2mg/L-IAA0.5mg/L为分化培养基;1/2MS+0.5mg/LNAA+IAA0.25mg/L为生根培养基进行培养。结果表明,幼根能产生少量愈伤组织,尚未分化出苗;子叶愈伤组织能分化出苗,但分化率低;下胚轴愈伤组织不仅可分化出苗(分化率为36%),而且可形成大量具有分化潜力的胚性愈伤组织,分化绿苗的频率可达67%。
Explants were hypocotyls , young roots , cotyledons of aseptic seedlings of blackseedcoat seame。Inducedmediumfor calliwas MSmediumwith2,4- D1.5mg/LandZ T0.5mg/L。Differentiation mediumforbudwas MSmediumwith6- BA2mg/LandIAA0.5 mg/L。Inducedmediumfor root was1/2MSmedium withNAA0.5mg/LandIAA0.25 mg/L;Theresultsindicated:afew calliformedfrom young rootshvitro,butthere werenobudsformedinculture。Somebudsweredifferentiatedfrom calliofcotyledonsCallifromhypocolylsnotonlydifferentiatedtosomebudsbutalsoformedalotofembryoniccalliwhichhaddifferentiation capacity. The differentiation ratio of green shoots could reach to 43%。 Hypoco1ylswerethebestexplantsinthecultureofblackseedcoatseameinvitro。
