摘要
目的探讨番茄红素联合α-生育酚对晶状体氧化损伤进行药物治疗的可行性。方法体外培养正常大鼠晶状体,按随机分组原则将大鼠晶状体分为以下8组:空白对照组(A),过氧化氢处理组(B),过氧化氢和不同浓度番茄红素(1μmol/L、5μmol/L、10μmol/L)处理组(C1、C2、C3),过氧化氢和不同浓度番茄红素(1μmol/l、5μmol/L、10μmol/L)联合α-生育酚处理组(D1、D2、D3)。紫外分光光度计测定晶状体组织的总抗氧化能力(T-AOC)、丙二醛(MDA)、谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的变化。结果培养24 h结束后,不同浓度番茄红素处理组及不同浓度番茄红素联合α-生育酚用药组SOD、CAT活性和GSH、T-AOC含量均显著高于过氧化氢处理组,MDA含量均显著低于过氧化氢处理组,经统计学分析,差异有显著统计学意义(P<0.01),不同浓度番茄红素处理组以5μmol/l抑制晶状体氧化损伤的效果最明显;联合应用α-生育酚可以增强番茄红素的作用。不同浓度番茄红素处理组与联合用药组各小组间两两比较,1μmol/L番茄红素联合α-生育酚处理组与5μmol/L番茄红素处理组间没有显著性差别,其余各组间差异均有显著统计学意义(P<0.01)。结论番茄红素可以抑制过氧化氢诱导的晶状体混浊,番茄红素联合α-生育酚抑制试验性白内障的形成是通过其抗氧化作用发挥作用,在α-生育酚存在的条件下可以增强番茄红素的作用,使低浓度的番茄红素发挥更好的作用。番茄红素联合α-生育酚有望成为预防或治疗白内障的新药。
Objective To explore the validity of lycopene in association withα-tocopherol on the treatment of hydrogen peroxide induced cataract. Methods Cultured normal rat lens in vitro were randomly divided into 8 groups:the control group, the group treated by hydrogen peroxide,the group treated by hydrogen peroxide and lycopene( 1 umoL/L, 5 umoL/L, 10 umoL/L), the group treated by hydrogen peroxide and lycopene( 1 umoL/L ,5 umoL/L,10 umoL/L) in association with α-tocopherol. The apoptosis ratio of lens on the time points of 24 hours were determined by flowcytometry. At the end of the incubation period, the lens were examined for biochemical parameters such as malondialdehyde (MDA) superoxide dismutase(SOD) catalase(CAT) glutathione (GSH) and total anfioxidiation ability(T-AOC) by spectrophotometrically. Results To compare with the control group, the level of T-AOC GSH and the activity of SOD CAT of the group treated by hydrogen peroxide reduced statistically;the level of MDA increased statistically (P 〈 0.01 ). Above mentioned biochemical parameters had altered statistically of the group treated by hydrogen peroxide and lycopene( 1 umoL/L,5 umoL/L,10 umoL/L) and of the group treated by hydrogen peroxide and lycopene ( 1 umoL/1,5 umoL/1,10 umoL/1 ) in association withα-tocopherol. Conclusion The lycopene can inhibit the turbidity of lens in vitro. Lycopene in association with α-tocopherol protects against experimental cataract development by virtue of its antioxidant properties. The lycopene in association with α-tocopherol can enhance antioxidiation ability of lycopene. Lycopene in association with α-toeopherol may be useful for prophylaxis or therapy against cataract.
