摘要
筛选出1株能分解纤维素的嗜碱性菌株,命名为XW12.均匀设计法优化摇瓶发酵产纤维素酶的条件,实验结果表明:最佳发酵条件为基质含蛋白胨0.57g,酵母粉0.76g,CMC0.56g,KH2PO40.0084g,酶活力可达45.54U/mL.对该菌株产生的纤维素酶粗酶液的酶学性质分析,结果表明:pH为8.0,50℃下表现出最大且稳定的酶活力,且Ca2+对酶反应有一定的促进作用.
One strain called XW12 capable of degrading cellulose was screened and named XW12. The optimal conditions of fermentation for producing CMCase were obtained by Uniform Design as follows= a medium containing peptone 0. 57 g, yeast extract 0.76 g, CMC 0. 56 g, KH2PO4 0. 008 4 g, and the enzyme activity was up to 45.54 U/mL. We also take some research for the characteristics of CMCase. The research shows that the optimum pH of crude CMCase of XW12 is 8.0, the optimum temperature 50 ℃, and it is stable in 50 ℃. Then the enzyme activity can be increasing by Ca^2+.
