雌激素及压力对髁突软骨细胞增殖与碱性磷酸酶活性联合效应的研究 被引量:8

摘要 目的:探讨雌激素及压力对髁突软骨细胞增殖和碱性磷酸酶活性的联合刺激效应。方法:体外培养髁突软骨细胞分为对照、90kPa/1h刺激、高、中、低浓度雌激素刺激及压力与各实验浓度雌激素联合刺激组,采用MTT法检测细胞增殖、酶动力学方法检测细胞ALP活性。结果:压力与雌激素都可促进MCCs的增殖与ALP活性,压力的存在并不影响外源性雌激素的促细胞增殖作用,但外源性雌激素可对压力的促增殖作用产生明显的抑制效应,双因素联合作用对ALP活性的促进作用最强。结论:雌激素与压力联合作用对髁突软骨细胞的增殖并不产生叠加效应,但对ALP活性显示出明显的叠加刺激效应。 Objective: To investigate the effects of mechanical pressure and estrogen on the proliferation and alkaline phosphatase (ALP) activity in rabbit mandibular condylar chondrocytes (MCCs). Methods: In vitro cultured MCC from two - week - old New Zealand rabbits were incubated and treated with single stimulation of controlled hydraulic pressure of 90kPa/1 h, 10^-6 mol/L, 10^-9 mol/L, 10^-12 mol/L 17β - estradiol ( E2 ), and combined stimulation of mechanical pressure and three different concentration of E2. ALP activity was determined by modified enzyme dynamical method and cell preliferation was studied with MTT assay. Results: Pure hydraulic pressure of 90kPa/1h or E2 caused increase in MCCs proliferation. Combined stimulation of mechanical stimulation and 10^-6 mol/L or 10^-9 mol/L 17 - β estradiol could impair the promotion effects of pressure to cell proliferation, with the higher concentration estrogen showing the even higher reverse effects. 10^-12mol/L 17 -β estradiol didn't influence the promotion effects of pressure to MCCs proliferation. As to ALP activity, the mechanical pressure and different concentration of estrogen all stimulated the ALP activity of MCCs. Under the combined stimulation of pressure and estrogen, ALP activity increased higher than either mechanical pressure group or the estrogen group. Conclusion: Both mechanical pressure and exogenous estrogen could stimulate the proliferation and ALP activity of MCCs. Mechanical pressure exists or not will not affect the promoting effects of estrogen to cell proliferation, while the exogenous estrogen evidently restrains the promoting effects of pressure to cell proliferation. The combined stimulation of mechanical pressure and estrogen exert the fortified stimulation to ALP activity of MCCs.
