摘要
目的:探讨壳聚糖-胶原支架在人牙周韧带细胞(PDLC)体外培养中的作用。方法:将体外培养PDLC接种于壳聚糖-胶原支架上,通过MTT法测定细胞增殖情况,通过半定量PCR检测细胞mRNA合成变化。结果:与传统的平板培养相比,PDLC在壳聚糖-胶原支架上的增殖及mRNA合成情况有显著性差异(P<0.05)。结论:该复合支架可以作为PDLC的载体;对PDLC的增殖及细胞外基质的合成均有较好地促进作用。
Objective: To study chitosan/collagen scaffold and observe the biological active of human periodontal ligament cell cultured on the scaffold in vitro. Methods: PDLC were seed on scaffold. Its proliferation was examined by MTT method and the effect of scaffold on mRNA synthesis was examined by semi - quantitive RT - PCR. Results: In contrast to control cultivation, on the proliferation and ECM mRNA synthesis of PDLC there had when cultured on the scaffolds. Conclusion: As a good carrier system, the complex scaffolds have significant effects on the proliferation of PDLC and ECM synthesis.
