摘要
目的:探讨上皮性卵巢癌患者外周血Th1/Th2细胞因子表达水平.方法:用刺激物刺激细胞,增加细胞内细胞因子的表达,再加入荧光标记的特异性抗细胞因子单克隆抗体,流式细胞仪检测特异性细胞因子表达水平.结果:上皮性卵巢癌患者Thl型细胞因子IL-2,IFN-γ表达水平依次为(9.47±3.56)ng/L,(10.12±2.51)ng/L,而正常对照组依次为(12.20±2.48)ng/L,(13.38±2.89)ng/L,差异均有统计学意义(P<0.05);而Th2型细胞因子IL-4,IL-10表达水平依次为(4.56±0.78)ng/L,(5.26±2.27)ng/L,而正常对照组依次为(1.24±0.39)ng/L,(1.46±0.52)ng/L,两组差异有统计学意义(P<0.05).结论:卵巢癌患者Thl/Th2失调,Th1型反应弱,Th2型反应优,可使肿瘤细胞发生免疫逃逸.
AIM: To detect the levels of Thl and Th2 type cytokines in peripheral blood of patients with epithelial ovarian cancer, METHODS: The peripheral blood lymphocytes were stimulated to enhance the expression of cytokines, Then specific fluorescent-labeling monoclonal antibody was added, The expression of the cytokines was determined by flow cytometry, RESULTS : The levels of IL-2 [ ( 9, 47 ± 3.56 ) ng/L] and IFN-γ[ ( 10.12 ± 2, 51 ) ng/L] produced by Thl cells were lower in patients with ovarian cancer than those in normal controls [ ( 12.20 ± 2.88), ( 13. 38 ± 2. 89) ng/L, respectively] and the difference was significant ( P 〈 0, 05 ). The levels of Th2 type cytokines, such as IL-4 and IL-10 were higher in patients with ovarian cancer [ (4.56 ±0.78), (5.26 ±2.27) ng/L, respectively]than those in normal controls [ ( 1.24 ± 0.39 ), ( 1.46 ± 0.52) ng/L. respectively ] with significant difference ( P 〈 0. 05 ), CONCLUSION : Th 1 / Th2 is unbalanced in ovarian cancer, and Thl type cytokines are inhibited, and Th2 type cytokines are relatively enhanced, so Th1/Th2 shifts to Th2, It may be the mechanism of tumor genesis and metastasis through immune escape from immunosurveillance.
