钨酸钠对脂肪细胞糖代谢的影响

摘要 目的:探讨钨酸钠对脂肪细胞葡萄糖代谢的影响及其机制。方法:3T3-L1前脂肪细胞诱导分化为成熟的脂肪细胞后,分别以不同浓度(0,150,300,500,700μmol/L)的钨酸钠处理48h后,用葡萄糖氧化酶法检测脂肪细胞对培养液中的葡萄糖消耗量,用2-脱氧-[3H]-D-葡萄糖摄入法检测葡萄糖转运率,用半定量RT-PCR观察葡萄糖转运子-4(glucose transport-4,GLUT-4)mRNA的表达。结果:150,300,500,700μmol/L钨酸钠能显著增加脂肪细胞葡萄糖消耗量和转运率,且呈剂量依赖效应。钨酸钠能增加GLUT-4mRNA的表达。结论:钨酸钠可能通过上调GLUT-4mRNA的表达而促进脂肪细胞糖代谢。 Objective To explore the effect of sodium tungstate on glucose metabolism in adipocytes and its mechanism. Methods After 3 T3-L1 preadipocytes were differentiated into adipocytes, these adipcytes were incubated with sodium tungstate (0, 150, 300, 500, and 700μmol/ L) for 48 h, and then glucose consumption of the adipocytes was detected by glucose-oxidase assay. Glucose transport was determined by the uptake of 2-deoxy-[^3H]-D-glucose, and the expression of glucose transport-4(GLUT-4) mRNA was identified by semi-quantitative RT-PCR. Results Sodium tungstate (150-700μmol/L) could significantly increase the glucose consumption and glucose transport with a concentration dependent-effect. Sodium tungstate could increase GLUT-4 mRNA expression. Conclusion Sodium tungstate can enhance the glucose metabolism of adipocytes by upregulating the expression of GLUT-4 mRNA.
