应用SSR标记技术鉴定马铃薯杂交实生种子的纯度 被引量:12

摘要 以昆明市农科所提供的2003年收获的B16、B19、B20及其亲本CIP004(♂)、CIP008(B16♀)、CIP009(B19♀)、CIP010(B20♀)为材料,利用9对SSR引物对其进行分析,筛选出适合用于马铃薯杂交实生种子纯度鉴定的SSR位点引物并初步测定其纯度。分析结果表明,STM002和STM009引物的扩增产物呈现双亲互补的特征谱带,易于区分杂交种和亲本自交系,并初步确定B、B和B纯度分别为86%、90%和85%。 In this study, Hybrid true potato seeds B16, B19 and B20 that they were harvested in 2003 by the In-titute of Kunming Agriculture Science and their parents CIP004( ♂ ) ,CIP008( B16 ♀ ) ,CIP009( B19 ♀ ) and CIP010(B20 ♀ )were used as experimental material to examine their purities via SSR. 9 SSR primer pairs were screened to search for the SSR loci that are suitable for testing the purity of hybrid true potato seed, The results are as follows : Purity of hybrid true potato seed was tested via SSR, we choosed that primers, which were STM 002 and STM 009, were fit for purity testing of hybrid true potato seed. We estimated that purity of B16 B19,B20 is respectively 86% ,90% ,85%.
