海马髓鞘碱性蛋白与中枢神经系统应变效应的实验研究 被引量:3

摘要 为探讨远离大脑的局部致伤条件下,脑组织和体液中髓鞘碱性蛋白(MBP)的活性变化,应用核酸斑点杂交和酶联免疫吸附法分别检测了犬双后肢低、高速投射物致伤后8h脑组织MBP-mRNA表达的变化和各时相点海马、丘脑下部、颞顶叶灰、白质和血浆、CSF-MBP含量的变化。MBP-mRNA表达:低速组下丘脑显著高于对照组(P<0.05),高速组海马、丘脑下部均非常显著高于对照组和低速组(P<0.01);低、高速组海马、丘脑下部、颞顶叶灰、白质和血浆、CSF的MBP含量均显著高于对照组和伤前(P<0.05),各脑区以海马、丘脑下部升高最显著。低、高速组血浆、CSF的MBP含量与其各脑区平均MBP含量均呈显著正相关(n=16,r分别为0.505和0.768,P分别<0.05和0.001)。说明远离大脑的局部致伤可致脑组织间接损伤,其损伤的发生与海马、丘脑下部的应激反应有关。 To investigate the activitic changes of myelin basic protein(MBP) in cerebral tissue, plasma and CSF following local injury far, from brain, spot hybridization and ELISA, we determined respectively the levels of MBPmRNA expression in every brain areas and the content of MBP in plasma, CSF, hippocampus, hypothalamus and temporoparietal of dogs with bilateral hind thigh injured by low, high velocity projectiles. The levels of MBPmRNA expression at 8th hour in hypothalamus following the bilateral hind thigh of the dogs injured by low velocity projectiles were significantly increased than control group(P<005), the levls of MBPmRNA expression in hypothalamus and hippocampus were very significantly increased in both low and high velocity projectiles injured groups than in preinjury and control group(P<001). The concentration of MBP in plasma, CSF,hippocampus, hypothalamus and temporoparietal were significantly increased in both low and high velocity projectiles injuryed groups than in preinjury and control group(P<005). The content of MBP in hippocampus and hypothalamus were the highest in all brain areas. The levels of MBP concentration in plasma or CSF were positively corvelated significantly with the mean MBP levels of all brain areas(r=0505 and 0768, respectively,n=16,P<005 and 0001, respectively). The local injury far from brain markedly induced the disturbance of CNS homestasis, which was related with the stress reaction of hippocampushypothalamus.
