摘要
探讨了全反式维甲酸(RA)对人肺癌A549细胞中尿激酶(uPA)及其受体(uPAR)基因调控作用。在细胞培养过程中用2.5μmol/L和10μmol/LRA处理A549细胞,两种浓度的RA均可抑制A549细胞生长,以10μmol/LRA作用明显;DNA-RNA斑点杂交检测显示,RA对A549细胞uPA及uPAR基因表达具有剂量依赖性调控作用,10μmol/LRA可显著抑制A549细胞中uPA及uPARmRNA表达,而2.5μmol/LRA则无明显效应。结果证明,一定浓度的RA可调节肿瘤细胞中uPA和uPARmRNA表达,RA对肿瘤细胞uPA和uPAR表达的下调作用有助于进一步阐明RA抑制肿瘤细胞浸润和转移的机制。
The effects of alltrans retinoic acid (RA) on the modulation of urokinase plasminogen activator (uPA) and its receptor (uPAR) mRNA were studied in human lung cancer A549 cell line. DNARNA dot blot hybridization assays indicated that the expression of uPA and uPAR mRNA had dosedependent regulating action by RA, and were inhibited obviously by 10 μmol/L RA. This finding suggested that uPA mRNA was regulated by RA at a certain concentration. The inhibitory effects of RA on uPA and uPAR gene expressions in this study may help to clarify the mechanism of RAinduced inhibition effect on tumor invasion and metastasis.
