摘要
目的探讨干扰素对骨肉瘤转移的调节作用,为临床应用干扰素防治骨肉瘤转移提供一定实验基础。方法采用人白细胞干扰素(IFNα)和免疫干扰素(IFNγ)对体外培养的人骨肉瘤细胞系OS732nm23H1蛋白的表达进行诱导调节,利用CelELISA半定量法检测其表达水平的变化。结果IFNα能显著上调OS732细胞nm23H1蛋白的表达水平(P<001)。结论IFNα可能在一定程度上抑制骨肉瘤细胞的转移潜能。
PURPOSE To explore the regulatory effects of IFN on invasive and metastatic process of osteosarcoma, and provide experimental evidence for clinical diagnosis and treatment of tumor metastasis.METHODS In this study, the changes of nm23H1 protein were halfquantitatively measured by CellELISA in human osteosarcoma cell line OS732 treated by IFNα and IFNγRESULTS The results demonstrated that IFNα has been shown to be capable of significantly increasing the expression of nm23H1 protein (P<0.01) in OS732 in a dosedependant manner, but the effects of IFNγ is not significant (P>005)CONCLUSIONS The findings suggest that IFNα may be able to suppress the metastatic potential of osteosarcoma and play a role in the treatment of patients with osteosarcoma.
