摘要
应用抗受体相关蛋白羧基端108个氨基酸多肽单克隆抗体F2(MonoclonalantibodyF2,McAbF2)进行被动型Heymann肾炎(PassiveHeymannnephritis,PHN)诱导试验。Western-Blot分析显示,McAbF2识别天然膜抗原Fx1A中分子量为44KDa的受体相关蛋白(Receptor-asociatedprotein,RAP),SD大鼠体内注射McAbF24天后肾皮质免疫组化和免疫电镜发现McAbF2在体内能识别肾小球上皮细胞表面抗原,并与之结合形成典型的PHN免疫复合物。表明在RAP羧基端108个氨基酸多肽上存在着Heymann肾炎病理性表型。
The experiment of inducing passive Heymann nephritis(PHN)with anti-COOH-terminal of RAP monoclonal antibody F2(McAb F2)could recognize the 44KDa RAP in the antigens of rat microvillar membranes(fx1A).4days after McAb F2 was injected into SD rats by vein,immunohistochemical and immunoelectron microscopy showed that McAb F2 could recognize and bind to the antigen on the surface of the glomerular epithelial cell,and form typical immune complexes of PHN in vivo.The result indicates that there is a nephritogenic epitope in the COOH-terminal 108 amino acids polypeptide of RAP.
