瘦素受体异构体ob-Ra和ob-Rb在胃腺癌组织中的表达 被引量:7

摘要 目的:研究瘦素受体异构体ob-Rao、b-Rb mRNA在胃腺癌组织及细胞中表达,探讨瘦素受体异构体ob-Ra、ob-Rb表达在胃腺癌发生、发展过程中作用和意义。方法:采用RT-PCR方法检测26例具有瘦素和瘦素受体蛋白双重表达的胃腺癌组织标本中瘦素受体异构体ob-Rao、b-Rb mRNA的表达情况,同时采用组织原位RT-PCR方法检测胃癌细胞中瘦素受体ob-Ra、ob-Rb mRNA的表达定位情况。结果:26例胃腺癌组织标本ob-Ra、ob-RbmRNA表达均为阳性,ob-Rao、b-Rb mRNA表达定位于胃腺癌细胞,表现为细胞质染成紫蓝色颗粒。结论:胃腺癌组织中有瘦素受体异构体ob-Ra、ob-Rb的表达,可以通过激活胃腺癌细胞上ob-Ra、ob-Rb受体下游的信号通路介导其促进肿瘤生长的生物学效应。 Objective: To investigate the expression of leptin receptor isoforms in gastric adenocarcinoma and its significance. Methods: The mRNA expression of leptin receptor isoforms ob-Ra and ob-Rb were assessed in 26 surgically resected specimens of gastric adenocarcinoma with dual expression of leptin and obJR using reverse transcriptional polymerase chain reaction (RT-PCR). The localiJ zation of ob-Ra and ob-Rb mRNA in gastric adenocarcinoma ceils were identified using in situ reverse transcriptional polymerase chain reaction (ISRT-PCR). Results: The expression of ob-Ra and ob-Rb mRNA were detected in all gastric adenocarcinoma specimens (26/26) with the localization of ob-Ra and ob-Rb mRNA in gastric adenocarcinoma cells. Conelusion: Leptin receptor isoforms ob-Ra and ob-Rb are expressed in gastric adenocarcinoma cells. Leptin may exert its effect via activating the downstream signal pathway of ob-Ra and/or ob-Rb in gastric adenocarcinoma.
