摘要
用多聚酶链反应—单链构象多态性分析(Single-strandconformationpolymorphismanalysisofpolymerasechainreaction,PCR-SSCP)以及地高辛标记探针斑点杂交技术对30例子宫颈鳞癌组织中Rb抑癌基因及HPV16E7癌基因进行分析。结果30例子宫颈鳞癌组织中未检出Rb基因突变或缺失;20例癌组织中检出HPV16E7DNA,阳性率为67%。提示HPV16E7癌基因与子宫颈鳞癌密切相关。子宫颈鳞癌中Rb基因结构异常并不常见,而且与HPV16E7基因的存在状况无明显相关性。
To clarify the association between Rb gene alteration and the presence of HPV E7 oncogene in the oncogenesis of cervical squmous cell carcinoma,30 primary cervical squmous cell carcinomas for the structure of Rb gene(exon 20)by PCRSSCP silver staining method were examined.The presence of HPV16E7 DNA sequences was also analyzed by dotblot hybridization using digoxigenin labelled HPV16 E7 probe.No point mutation or loss of hetrozygosity in the Rb susceptibility genes was detected in any of 30 tumor specimens.HPV E7 DNA were found in 67%(20/30)of the tumors The results suggest that cervical squamous cell carcinoma is associated with HPV16 E7 oncogene.In Contrast to data derived from cultured cervical carcinoma cell lines,Rb gene alteration is infraquent in clinical samples of cervical carcinomas irrespective of the presence or absence of HPV E7 oncogene.
