高脂膳食对SR-AⅠ/Ⅱ基因敲除小鼠白色脂肪组织中UCP2 mRNA表达的影响

摘要 目的:观察SR-AⅠ/Ⅱ基因敲除(SR-AⅠ/Ⅱ-/-)小鼠白色脂肪组织中UCP2 mRNA的表达,探讨其与SR-AⅠ/Ⅱ-/-小鼠肥胖易感的关系。方法:应用荧光定量RT-PCR检测小鼠附睾周白色脂肪组织中UCP2 mRNA的表达;图像分析法评估附睾周脂肪细胞形态学变化;酶法检测甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)血脂水平。结果:经12周高脂饲料喂养,SR-AⅠ/Ⅱ-/-小鼠的体重、血脂(TG、TC、LDL-C)水平、附睾周脂肪垫重量、脂肪细胞面积和直径均大于野生(SR-AⅠ/Ⅱ+/+)对照小鼠(P<0.01);白色脂肪组织UCP2 mRNA表达量低于SR-AⅠ/Ⅱ+/+小鼠(P<0.01)。结论:SR-AⅠ/Ⅱ-/-小鼠对膳食诱导肥胖(diet-induced obesity,DIO)易感可能与其白色脂肪组织UCP2 mRNA表达异常有关。 AIM: To observe the relationship between UCP2 mRNA expression in white adipose tissue and diet - induced obesity in SR - A Ⅰ/Ⅱ gene knock - out ( SR - AⅠ/Ⅱ -/- ) mice. METHODS : Fluorescent quantitative RT - PCR was used to detect UCP2 mRNA expression in mice epididymal white adipose tissue. The cellular morphological changes were analyzed by using image analysis. Serum TG, TC and LDL - C concentrations were measured by enzymatic determination. RESULTS: After fed with high fat diet for 12 weeks, average body weight of SR - A Ⅰ/Ⅱ-/- mice was much higher than that of wild type ( SR - A Ⅰ/Ⅱ -/- ) control mice ( P 〈 0. 01 ), as well as the serum lipid ( TG, TC, LDL - C) levels, epididymal fat pad weight, adipocyte area and diameter. UCP2 mRNA expression in white adipose tissue in SR - A Ⅰ/Ⅱ-/- mice was significant lower than that in SR - A Ⅰ/Ⅱ-/- control, mice (P 〈 0. 01 ). CONCLUSION: Abnormal UCP2 mRNA expression may be associated with SR - A Ⅰ/Ⅱ -/- mice susceptive to diet - induced obesity.
