丙戊酸钠诱导K562细胞周期阻滞和凋亡并上调p^21WAF1基因mRNA的表达 被引量:6

摘要 目的:探讨丙戊酸钠(VPA)对慢性粒细胞白血病急变期细胞株(K562细胞)细胞周期和凋亡的影响及其可能机制。方法:利用流式细胞仪检测药物处理后的细胞周期和细胞凋亡情况;利用RT-PCR方法检测p21WAF1基因的mRNA的表达。结果:VPA引起K562细胞凋亡增加[(11.47±0.25)%vs(4.77±0.40)%,(P<0.05)]和细胞周期阻滞在G0/G1期[(82.30±9.41)%vs(40.13±2.12)%,(P<0.05)]。同时处理后的细胞中p21WAF1基因mRNA的表达增加[(1.65±0.91)vs(0.25±0.04),P<0.05]。结论:VPA可能通过上调p21WAF1基因,导致K562细胞周期阻滞并引起细胞凋亡。 AIM: To investigate the effects of sodium valproate (VPA) on the cell cycle and apoptosis of chronic myeloid leukemia cell line K562, and to explore the possible mechanisms. METHODS: K562 cells were treated with VPA. Cell cycle and apoptosis were analyzed by flow cytometry. The expression of P^21WAF1 mRNA was detected by RT - PCR. RESULTS : After treatment with VPA, cell cycle was arrested obviously at G0/G1 phase [ ( 82. 30 ± 9.41 ) % vs (40. 13 ± 2. 12)% , P 〈 0. 05 ]. The apoptotic rate was significantly higher in the cells treated with VPA than that in untreated cells [ ( 11.47% ± 0. 25% ) vs (4. 77% ± 0. 40% ), P 〈 0. 05 ]. The level of P^21WAF1 mRNA was increased [ ( 1.65 ±0. 91) vs (0. 25 ± 0.04), P 〈 0. 05 ]. CONCLUSION: VPA induces elevated expression of P^21WAF1 mRNA in K562 cells, resulting in G0/G1 phase arrest and apoptosis in vitro.
