热疗与紫杉醇联合应用对H446细胞生长和MMP-2mRNA表达的影响 被引量:1

摘要 目的:探讨单纯热疗、单纯化疗以及热化疗联合应用对H446细胞生长及MMP-2mRNA表达的影响。方法:以不同温度(39℃、40℃、41℃、42℃、43℃和44℃)、不同剂量紫杉醇(30μg/L、60μg/L、120μg/L、240μg/L和480μg/L)及热化疗联合的方法处理H446细胞,同时设对照组(0μg/L紫杉醇,37℃),每组又分为3个热疗时间(30min、60min和90min),观察热疗、化疗和热化疗联合对H446细胞生长抑制率的影响;另取H446细胞,分为单纯热疗组(43℃,90min),单纯紫杉醇化疗组(60μg/L、120μg/L和240μg/L),43℃热疗90min联合紫杉醇组(60μg/L、120μg/L和240μg/L),对照组(0μg/L、37℃),采用RT-PCR检测各组细胞MMP-2mRNA的表达。结果与结论:单纯热疗与紫杉醇均可抑制H446细胞的生长,热疗不同程度地增强了紫杉醇对H446细胞的增殖抑制率,热化疗联合对H446细胞生长抑制的最优组合是43℃热疗90min联合120μg/L紫杉醇化疗,在此条件下,H446细胞MMP-2mRNA的表达明显降低,提示热化疗联合对H446细胞增殖的抑制作用可能是通过降低MMP-2mRNA的表达,从而抑制肿瘤细胞的侵袭和转移而实现。 Aim:To investigate the effects of thermotherapy,chemotherapy (with paclitaxel) and thermochemotherapy on cell proliferation and MMP-2 mRNA expression of Hgg6 cells. Methods:Different temperatures (39 ℃ ,40 ℃ ,41 ℃ ,42 ℃ ,43 ℃ and 44 ℃ ) ,different time (30 min,60 min and 90 min) of hypcrthermia, and different doses(30 μg/L,60 μg/ L, 120 μg/L,240 μg/L and 480 μg/L) of paclitaxel were grouped respectively. MMT assay was adopted to measure cell proliferation and the inhibition rates. Semi-quantitive RT-PCR was applied to determine the expression of MMP-2 mRNA. Results and Conclusions: Paclitaxel and thermotherapy both could inhibit the growth of H446 cells and thermotherapy enhanced the inhibitive effects of paclitaxel on the H446 cell proliferation. The best group of temperature,time and paclitax- el concentration is thermotherapy at 43 ℃ for 90 min and 120 μg/L paclitaxel. Thermochemotherapy could evidently decrease the expression of MMP-2 mRNA.
