芪玄益心胶囊对大鼠心肌NF-κBp50mRNA IκBαamRNA的影响

摘要 目的:评价芪玄益心胶囊对冠脉结扎大鼠心肌梗塞的治疗作用,并探讨其机理。方法:建立大鼠冠脉结扎心肌梗塞模型,将模型大鼠随机分为麝香保心丸组、芪玄益心胶囊高、中、低剂量组、模型组,并设空白对照组。观察各组心肌梗塞率和心肌组织核转录因子-κBp50(NF-κBp50)其抑制蛋白α(IκBα)的表达。结果:模型组及各治疗组与空白组比较心肌梗塞率、NF-κBp50mRNA、IκBαmRNA表达均显著增高(P<0.05)。芪玄益心胶囊各剂量组与模型组比较心肌梗塞率、NF-κBp50mRNA、IκBαmRNA均明显降低(P<0.05),以高剂量组变化最显著,高剂量组在降低心肌梗塞率、NF-κBp50mRNA、IκBαmRNA表达方面均优于麝香保心丸组(P<0.05)。结论:芪玄益心胶囊能够降低心肌梗塞率,并推测芪玄益心胶囊抗心肌梗塞的作用与其抑制NF-κBp50mRNA、IκBαmRNA表达相关。 Objective :To evaluate the therapeffllc effect of capsule Qixuan Yixin on myocardial infarction in coronary ligated rats. Methods : To ligate coronary of rats to duplicate myocardial infarctioil model. The rats were randomized into Shexiang Baoxin pill group, high ,middle and 10w dose of capsule Qixuan Yixin group,Plaeebo group and control group. Observe the rate of myocardial infarct ion and the expression of mRNAof NF- κBp50 and IκBα in myocardial. Results : Compared with control group, the rate of myocardial infarction and the expression of mRNA of NF- κBp50 and IκBα in myocardial were higher in PIacebo group and each therapeutie group( P 〈 0. 05 ). The rate of myocardial infarction and the expression of mRNA of NF- κBp50 and IκBα in myocardial were lower in each dose of capsule Qixuan Yixin ( P 〈 0.05). With high dose group showed the most obvious effect, which was superior to Shexiang Baoxing Pill( P 〈 0. 05). Conclusion :Capsule Qixxuan Yixin could lower the rate of myocardial infarction. Speculate that the ailti - infaretion effect was associated with the decreased expression of mRNA of NF- κBp50 and IκBα .
