摘要
以含5%甲醇和0.02mol/L邻苯二甲酸氢钾的缓冲液(pH 3.52)为流动相,采用安培型电化学检测器,实现了左旋多巴、甲基多巴、多巴胺和去甲肾上腺素在Shim-pack CLC-ODS(5μm,150 mm×60mmI.D)柱上的完全分离及E=+0.6 V电压下于玻碳电极上的同时检测。该法线性范围达2个数量级,相关系数大于0.9997,最小检出限为 28~94pg,实验回收率为 84.9%~91.3%。该法快速、灵敏、准确,适用于体液中痕量代谢药物的分析,可用于临床监测。
Trace amount of levodopa, methyldopa, dopamine, and noradrenaline can be determined simultaneously by RP-HPLC on a column of Shim-pack CLC ODS C18, by using a mobile phase of 5% methanol solution and 95% 0.02 mol/L potassium biphthalate buffer(pH 3.52) and an electrochemical detector, at 0.06 V with a detection Limit of 28.0 - 94.0 pg, a linear range of 0.12 - 100 ng, a recovery range of 84.7% - 91.3%, and a correlation coefficient of higher than 0.9997.
