摘要
目的探讨自然流产患者外周血单个核细胞和胎盘组织转化生长因子-β1(TGF-β1)的异常表达。方法采用逆转录-聚合酶链反应(RT-PCR)技术,半定量检测30例早期自然流产患者、25例正常妊娠妇女、25例正常未妊娠妇女外周血单个核细胞内TGF-β1mRNA表达;采用免疫组织化学技术检测绒毛和蜕膜组织内TGF-β1表达强度。结果早期自然流产患者、正常妊娠妇女、正常未妊娠妇女3组研究对象外周血单个核细胞表达TGF-β1mRNA相对含量分别为21.5±9.4%、95.7±12.9%、87.0±13.8%,组间比较均有显著性差异(P<0.05);自然流产组合体滋养层细胞表达TGF-β1显著低于人工流产组(P<0.01)。结论正常早期妊娠妇女外周血单个核细胞TGF-β1mRNA表达水平较正常未妊娠妇女显著升高,提示TGF-β1对妊娠维护起重要作用;早期自然流产患者外周血单个核细胞TGF-β1mRNA表达水平及绒毛合体滋养细胞层表达TGF-β强度均较正常妊娠妇女显著下降,其低水平表达可能与自然流产相关。
Objective: To investigate the aberrant transforming growth factor betal mRNA expression in periphral blood mononu- clear cells (PBMCs) and placental tissues during early spontaneous abortion. Methods: We measured the TGF -β1 mRNA expres- sions in PBMCs from 30 early spontaneous aborters, 25 normal early pregnant women, and 25 normal women without pregnancy using semi - quatitative transcriptional - polymerase chain reaction ( RT - PCR) technique. We also identified TGF -β1 proteins in placental tissues using immunohistochemistry. Results: We observed the levels of TGF -β1 mRNA from the above three groups were 21.5 ±9.4%, 95.7 ± 12.9%, 87.0 ± 13.8%, respectively. Significantly differences were found among groups ( P 〈 0. 05 ) . As for the intensity of the immunohistochemistry signal, TGF -β1 in syncytiotrophoblastic cells of aborters was markedly lower than that of normal early pregnant women ( P 〈 0. 01 ) . Conclusion: The levels of TGF -β1 mRNA expression in PBMCs of normal early pregnant women are significantly higher than those of nomal non - pregnant women. This indicates that TGF -β1 may Contribute to the mainte- nance of pregnancy. The levels of TGF -β1 mRNA expression in PBMCs and TGF -β1 in syncytiotrophoblastie cells of aborters are sig- nificantly lower than those of the normal early pregnant women. The abnormal expression of TGF -β1 may be associated with pregnancy failure.
