摘要
以爱玉子的叶片为材料,对CTAB分离总DNA的方法进行了改进,即在提取液中加入β-巯基乙醇、PVP,将CTAB的浓度提高到3%,用氯仿-异戊醇取代酚进行抽提。结果表明,改进后的方法可有效地去除杂质,防止材料褐化,获得的DNA样品OD260nm/OD280nm比值为1.8左右,质量好且产率较高,能完全满足SRAP分析及限制性内切酶酶切的要求。
This paper explored an improved CTAB method to extract high-quality genomic DNA from the leaves of Ficus awkeotsang Makino, which was the application of β-SH (mercapto) ethanol and PVP, and increasing the concentration of CTAB up to 3 %, and substituting chloroform-isoamylol for phenol. The research results showed that this modified method can effectively elimi nate impurity and prevent DNA samples from turning brown. High-quality and high-yield genomic DNA with OD260nm/OD280nm approximate 1.8 was obtained, and it was fully suitable for SRAP analysis and restriction enzyme digestion.
