鼻咽非角化性癌miRNAs的差异表达及意义 被引量:4

摘要 目的分析鼻咽非角化性癌和癌旁慢性炎症组织的miRNAs差异表达谱。方法显微切割鼻咽非角化性癌及癌旁慢性炎症组织,提取RNA,分离小分子RNA,并用miRNAs芯片制作的微阵列进行杂交,杂交后通过芯片扫描和数据分析获得鼻咽非角化性癌miRNAs差异表达谱。结果鼻咽非角化性癌中表达上调的6个miR-NAs中,其差异表达在2倍以上的有2个miRNAs;表达下调的7个miRNAs中,其差异表达在2倍以上的有1个miRNAs。结论鼻咽非角化性癌中存在差异表达的miRNAs,包括miR-203、miR-503、miR-424、miR-141、miR-148α、miR-25、miR-195和miR-15α等,提示这些miRNAs可能与鼻咽非角化性癌发生发展有关,可作为鼻咽非角化性癌候选分子标记物,为进一步研究鼻咽非角化性癌的标记物提供帮助。 Objective To test and compare miRNAs expression in nasopharyngeal carcinoma and paracancerous chronic nasopharyngitis tissues using gene-chip miRNAs, and to analyze the expression profile of nasopharyngeal non-keratinizing carcinoma. Methods nasopharyngeal nonkeratinizing carcinoma and paracancerous chronic nasopharyngitis tissues were mici'odissected and miRNA was extracted. And then microarray hybridization was performed by gene-chip miRNAs. Finally, hybridization results were analyzed. Results Six miRNAs were up-regulated in nasopharyngeal non-keratinizing carcinoma compared with paracancerous tissues,while 7 miRNAs downregulated. Two of the 6 up-regulated and 1 of the 7 down-regulated miRNAs had a differential expression of 2 times and upwards. Conclusion Differentially-expressed miRNAs exist in nasopharyngeal non-keratinizing carcinoma, including miR-203, miR-503, miR-424, miR-141, miR- 148α,miR-25,miR-195 and miR-15α,indicating that they might be related to nasopharyngeal non- keratinizing carcinoma and can be taken as its potential molecular marker, thus a further study of the potential markers of nasopharyngeal non-keratinizing carcinoma can be made.
