摘要
目的:观察二巯基乙醇(2-ME)对大鼠骨肉瘤细胞增殖的诱导作用。方法:将含10%小牛血清的RPMI1640完全培养基分装成六瓶,每瓶100ml,一瓶不加2-ME作为对照组Ⅰ,其余五瓶分别加入不同剂量2-ME作为实验组,再设一个含10%胎牛血清的完全培养基一瓶作对照组Ⅱ,分别在96孔板中培养大鼠骨肉瘤UMR106细胞,24h、48h和72h后,观察细胞生长状态,采用MTT法检测细胞增殖情况。结果:对照组Ⅰ生长缓慢,对照组Ⅱ生长状态良好,分裂增殖相较多。实验组随着2-ME浓度的增加,细胞生长的速度逐渐增快。其中0.3μl组生长速度适中,细胞生长状态与对照组Ⅱ相似。结论:二巯基乙醇有明显诱导大鼠骨肉瘤细胞增殖的作用。
Objective:To explore the cell proliferation induction of rats' osteosarcoma cells treated by different concentration of 2-mereaptoethanol(2-ME).Methods:Five experimental groups were established and cultured with the RPMI1640 complete medium containing 10% calf serum with different concentration of 2-ME;control groupⅠ,with the RPMI1640 complete medium containing 10% calf serum without 2-ME;and control groupⅡ,with the RPMI1640 complete medium containing 10% fetal calf serum.The culture solutions of different groups were used to culture the rats' osteosarcoma UMR106 in 96-hole plate.The cell growth status was observed by microscope.The cell proliferation was detected by MTT assay after treated 24,48 and 72 hours.Results:The cells in the control groupⅠgrew slowly,but the cells in the control groupⅡgrew healthily.In the experimental groups,the cell proliferation increased with the concentration increasing.Conclusion:2-mereaptoethanol plays an important role in the induction of proliferation of rats' osteosarcoma cells.
