摘要
采用RT-PCR从猪外周血液淋巴细胞中克隆出猪γ-干扰素基因cDNA,构建了重组酵母表达载体(pPICZα-PoIFN-γ),并在巴斯德毕赤酵母X33株中实现了高效分泌表达,经SDS-PAGE和Western-blot证实猪γ-干扰素分子量为18 kD,细胞培养试验结果表明,重组猪γ-干扰素能抑制水泡性口炎病毒(VSV)对猪肾传代细胞(PK-15)的攻击;而在动物试验中,重组猪γ-干扰素可明显提高猪伪狂犬疫苗的免疫效果。
Porcine interferon-gamma(PoIFN-γ) cDNA was cloned by RT-PCR from porcine lymphocytes in peripheral blood. The recombinant plasmid (pPICZα-PoIFN-γ) encoding PoIFN-γ gene was constructed and transferred into Pichia pastoris X33 strain. A high level expression of PoIFN-γ in the recombinant Pichia pastoris was detected by SDS-PAGE analysis and Western-blot assay. The recombinant PoIFN-γ can protect the porcine kidney cells (PK-15) from the attack of vesicular stomatitis virus in vitro and significantly enhanced the immune response of pseudorabies virus vaccine in pigs.
