鲍曼不动杆菌耐药程度与其主动外排泵蛋白的相关性研究 被引量:3

Study on Correlation between Antibiotic Resistance and Active Efflux Transporters in Acinetobacter Baumannii

摘要 目的:探讨鲍曼不动杆菌耐药程度与其主动外排泵蛋白的相关性。方法:首先用纸片扩散法检测64株临床鲍曼不动杆菌对8种抗菌药物的敏感性;将其分为A组(0~2种抗生素耐药)、B组(对3~5种抗生素耐药)和C组(对6~8种抗生素耐药);检测64株临床鲍曼不动杆菌对罗丹明6G的外排情况,筛选出罗丹明6G外排明显增加的菌株;并用逆转录-聚合酶链反应(RT-PCR)方法检测主动外排泵基因AdeABC的表达水平。结果:64株鲍曼不动杆菌中有4株对0~2种抗生素耐药(A组),对3~5种抗生素耐药的有33株(B组),对6~8种抗生素耐药的有27株(C组);多重耐药组鲍曼不动杆菌罗丹6G外排明显增高,外排程度A组<B组<C组;罗丹明6G外排增强的多重耐药不动杆菌主动外排泵基因AdeABC的表达水平显著增高。结论:鲍曼不动杆菌的耐药现象严峻;多重耐药性与药物主动外排增强有关;AdeABC基因的过度表达是鲍曼不动杆菌药物外排增加的重要原因。 Objective: To investigate the function and gene expression of active effiux transporters in Acinetobacter baumannii. Methods: At first, the susceptibilities of 64 strains clinical Acinetobacter baumannii to 8 kinds of antibiotic agents were tested by disc dif- fusion method, then these strains were divided into three groups based on the susceptibilities: group A (resistant to 0-2 antibiotics), group B (resistant to 3-5 antibiotic) and group C (resistant to 6-8 antibiotics). The effiux of Rhodamine 6G in these 64 strains of clinical Acinetobacter baumannii was tested and identified, the strains with high increased effiux of Rhodamine 6G were screened out; Reverse transcriptase polymerase chain reaction (RT-PCR) was used to measure the mRNA expression levels of active effiux gene AdeABC. Results: Among the 64 strains, there were 4 strains resistant to 0-2 antibiotics (groupA), 33 resistant to 3-5 antibiotics (groupB)and 27 resistant to 6-8 antibiotics (groupC). The effiux of Rhodamine 6G was significantly increased in some multiple resistant strains, and the level ofadeABC were also significantly increased. Conclusion: The antibiotic resistance is very serious in Acinetobacter baumannii. The multiple resistance ofAcinetobacter baumannii is related to the active effiux ability. The overexpression of AdeABC gene induces the in- creased activity of active effiux in Acinetobacter baumannii.
