胃癌AGS细胞内pH值的测定及酸性环境对其增殖和凋亡的影响 被引量:2

摘要 目的:测定胃癌AGS细胞内pH(intracellular pH,pHi),探讨细胞外酸性环境对AGS细胞增殖和凋亡的影响。方法:采用离子成像分析系统测定AGS细胞的pHi,MTT及流式细胞术分别检测培养环境pH值为6.0时AGS细胞的活力、细胞周期和凋亡。结果:AGS细胞内pH明显碱化,在pH值为6.0的酸性环境培养时,细胞活力在12小时内明显抑制,细胞停滞在S和G2/M期,细胞凋亡不明显。结论:胃癌细胞AGS明显碱化,pH值为6.0的酸性环境使细胞数量减少和活力下降,其机制是使细胞出现增殖抑制而与凋亡无关。 Objective: To measure the intracellular pH (pHi) of the gastric cancer AGS cell and investigate the effect of acidic extracellular environment on the proliferation and apoptosis of AGS cell. Methods: The pHi of the AGS cells was detected with a fluorescent probe, BCECF, through an ion imaging analysis system. The cell viability was determined with MTT method and the proliferation and apoptosis of the cells were studied with flow cytometry. Results: The pHi of AGS cells was obviously alkalified to about 8.60, The viability and the proliferation of the AGS cells were inhibited remarkably and the cells were arrested at S and G2/M phase, No significant changes in cell apoptosis were observed. Conclusions: The gastric AGS cell was significantly alkalified. The number and the viability of AGS cells were decreased by culturing the cells at pH 6.0 condition,which is correlated with the inhibition of cell proliferation but not with cell apoptosis.
