摘要
研究了在培养液中添加细胞因子和不同激素对山羊卵母细胞体外成熟和胚胎体外发育的影响。结果表明:添加不同浓度激素组合能促进早期胚胎的发育(P<0.05),1μg/ml雌二醇(E2)、200μg/ml促黄体素(LH)和10μg/ml垂体促卵泡素(FSH)的浓度组合效果最好(P<0.05);胶质细胞源性神经营养因子(GDNF)、白血病抑制因子(LIF)、干细胞因子(SCF)对山羊卵母细胞体外成熟和胚胎体外发育有极显著影响(P<0.01),但不同浓度间无显著影响(P>0.05),添加10 IU/ml LIF、15 IU/ml GDNF和15 IU/ml SCF对山羊胚胎体外发育最好;GDNF+LIF+SCF组和GDNF+LIF组卵母细胞成熟率极显著高于LIF+SCF和GDNF+SCF组(P<0.01),GDNF+SCF和LIF+SCF组间对卵母细胞体外成熟无差异(P>0.05),早期胚胎发育上GDNF+LIF+SCF组高于GDNF+LIF和LIF+SCF组,也高于GDNF+SCF组,均能提高桑椹胚率,但试验组间差异不显著(P>0.05)。可见GDNF、LIF和SCF能提高卵母细胞的成熟率和桑椹胚率,LIF和GDNF对卵母细胞体外成熟有协同作用。
Effects of cytokine and hormone on in vitro maturation of oocyte and early embryo development of goat were studied. Different hormone combinations significantly improved early embryo development (P 〈 0. 05 ) , with 1μg/ml E2, 200 μg/ml luteinizing hormone (LH) and 10 μg/ml folicle-stimulating hormone (FSH) being the best combination. There was significant difference (P 〈 0. 01 ) between control group and experimental group (cytokine factor added) in oo-ytes maturation and marulae formation rate ( P 〈 0. 01 ), but no difference between cytokine solutions ( P 〉 0. 05 ). 10 IU/ml leukemia inhibitory factor (LIF), 15 IU/ml glial-cell-line-derived neurotrophic factor (GDNF) and 15 IU/ml stem cell factor (SCF) favored the early embryos development in goat. Maturation rate of goat oocytes was increased more in the groups with GDNF + LIF + SCF or GDNF + LIF than the groups with GDNF + SCF or LIF + SCF(P 〈0. 01 ). The results indicated that GDNF, LIF and SCF improved oocytes maturation and the marulae formation rate in goat, and LIF and GDNF had coeffects on in vitro maturation of goat oocytes.
